Now this is fine for the writer, but production needs to know how many scenes are in the script and which of those scenes are self-contained. The script is ready? Time to start breaking it down!īelow is an example of a Scene Heading from a writer’s draft: – Rachel Noll, Producer of The Storyteller. We wanted to get this stuff locked down early so we could start booking travel for our actors. ![]() This happened in January – 5 months out from the shoot. Once we had a general idea of what our locations were and where they were located, and had secured our cast and confirmed they had no conflicts with the shoot dates, we sat down with our 1st AD to break down and schedule the script. So how does a filmmaker know when their script is ready for breakdown? Now let’s be honest, a script is never truly finished. We waited until we were pretty certain there would be no more major changes – because we didn’t want to lock in the shoot dates until we were sure no locations would be changing dramatically. It won’t do you any good to start planning your production if the script is still in the collaborative stage. ![]() The first step in the script breakdown process is to have a script that you feel is ready to be broken down. This should not be confused with a casting breakdown, which is used when finding your actors. How does one find all those elements? Simple: They are in your script.Ī Script Breakdown is when the producer and/or the first assistant director take the locked script and go through and mark the occurrence of every element. To create the best schedule, you must account for every element in your story. The schedule provides a timeline for your film and allows you to fully plan how your budget is going to be divided. It includes when locations need to be secured, crew hired, when and where wardrobe and props are needed. A schedule isn’t limited to when you’re going to shoot the film. A key part of the pre-production process is building a schedule. ![]() 8/1/2023 0 Comments Scoop away![]() ![]() Scooping out clumps of urine and feces is something done regularly, every other day at minimum, but often daily or even twice daily if you have a cat in a small apartment and want to keep litter box smell under control. If you’re a clumping cat litter user (and about 60% of kitty parents are), there are two ways to clean the litter box: the scoop and the change. Two Types of Litter Box Cleaning: the Scoop and the Change In this case, you won’t have to clean the individual boxes any more often, you’ll just have two or more boxes to maintain. If you have more than one cat, you’ll want to have more than one litter box, too, so this helps distribute the urine and feces between the boxes. While this is true for the most part, some litters are designed for multiple-cat households and have strong odor-controlling ingredients to help prolong the time between cleaning. It stands to reason that the more cats you have, the more urine and feces will accumulate in the litter boxes and the more often you’ll have to clean them. Litter Box Maintenance Variable 2: Number of Cats Non-clumping litters cannot be scooped, so to clean them you must change out all of the litter each time.Ī litter that contains odor-controlling ingredients will keep down the smell and extend how often you will need to scoop the box or change out all of the litter. If you use non-clumping litter, urine will be absorbed by the clay or other ingredients such as corn or wood, with some falling to the bottom of the cat box under the litter.Ĭlumping litters need changing less frequently because you can use a litter scoop to remove the coated clumps of cat pee and poop. ![]() Clumping litter absorbs cat urine and forms hard clumps that can be scooped from the box, leaving the unsoiled litter behind. The type of litter you choose makes a difference in how often you’ll need to clean the cat box. Litter Box Cleaning Variable 1: Type of Litter How you clean and maintain a litter box depends on your situation. While you won’t have to pick up poo in a plastic bag after every deposit like a dog owner, you will have to adopt a maintenance routine of scooping out clumps of urine and feces and giving the litter box a thorough change and cleaning regularly. But since kitty’s toilet is inside, you do have the job of cleaning the litter box. Adopting a Good Litter Box Cleaning RoutineĪs a cat mom or dad, you’re spared the chilly mornings and rainy or snowy nights when pupper parents are out walking their dog and hoping she’ll “do her business” so they can get back inside where it’s warm and dry. ![]() 8/1/2023 0 Comments 3d model of fibrin jmol![]() During fibrinolysis, plasmin initially cleaves the αC regions, and then cleaves the three polypeptide chains connecting the central (E) and end (D) regions ( Fig. ![]() In particular, the αC regions contain lysine-dependent tPA- and plasminogen-binding sites (K d = 16–33 nM) within residues Aα392–610. Cryptic plasminogen and plasmin binding sites in fibrinogen and fibrin monomers are exposed by fibrin monomer polymerization. Following thrombin-mediated cleavage of fibrinopeptides A and B from the N-termini of the Aα- and Bβ-chains, respectively, fibrin monomers polymerize into half-staggered, double-stranded protofibrils that bundle into fibers via interactions of the αC regions. The fibrinogen molecule is comprised of three pairs of distinct peptide chains: two Aα-chains, two Bβ-chains, and two γ-chains, which are interlinked by disulfide bridges ( Fig. Plasmin is the activated form of plasminogen and is produced either in the blood stream by urokinase-type plasminogen activator (uPA), or on the fibrin surface, where bound plasmin and tissue-type plasminogen activator (tPA) are protected from their respective inhibitors, α 2-antiplasmin and plasminogen-activator-inhibitor 1. The primary enzyme mediating fibrinolysis is the serine protease plasmin, which cleaves the fibrin molecule at specific sites. When the hemostatic role of the clot is fulfilled, it is removed via the fibrinolytic system. Blood clots contain an interconnected web of fibrin and blood cells, including platelets, that mediate clot retraction. Inappropriate clot formation within blood vessels (thrombosis) leads to ischemia and tissue loss. Blood clots must be strong enough to limit extravascular blood flow following injury, yet dissolve appropriately during wound healing. The hemostatic system must strike a balance between hemorrhage and vessel occlusion. These results highlight how subtle differences in the diameter and prestrain of fibers could lead to dramatically different lytic susceptibilities.Ĭitation: Bucay I, O’Brien ET III, Wulfe SD, Superfine R, Wolberg AS, Falvo MR, et al. Using polymer physics- and continuum mechanics-based mathematical models, we show that fibers polymerize in a strained state and that thicker fibers lose their prestrain more rapidly than thinner fibers during lysis, which may explain why thick fibers elongate and thin fibers lyse. Because lysis rates were greatly reduced in elongated fibers, we hypothesize that plasmin activity depends on fiber strain. Thinner fibers were more likely to lyse, while fibers greater than 200 ± 30 nm in diameter were more likely to elongate. Thrombin and plasmin dose-response experiments showed that the elongation behavior was independent of plasmin concentration, but was instead dependent on the concentration of thrombin used during fiber polymerization, which correlated inversely with fiber diameter. ![]() We found that during lysis 64 ± 6% of fibers were transected at one point, but 29 ± 3% of fibers increase in length rather than dissolving or being transected. Using an inverted optical microscope and fluorescently-labeled fibers suspended between micropatterned ridges, we have directly measured the lysis of individual fibrin fibers. To understand blood clot dissolution, the influence of clot structure and fiber properties must be separated from the effects of enzyme kinetics and perfusion rates into clots. Fibrin fibers form the structural backbone of blood clots fibrinolysis is the process in which plasmin digests fibrin fibers, effectively regulating the size and duration of a clot. ![]() |
